不同体况对妊娠初期母牛阴道菌群的影响

摘要:
为研究不同体况对妊娠初期母牛阴道菌群的影响,采集相同放牧条件下不同体况妊娠初期西门塔尔母牛18 头(每组6头,偏瘦组(TC),标准组(MC)和肥胖组(FC))阴道分泌物,利用16S rRNA测序技术对其阴道菌群多样性进行分析。结果显示:18 份样品平均每份样品测得70 567条序列,经过质控平均得到68 309条序列,质控有效率达96.84%;通过聚类共获得2 677 个操作分类单元(Operational taxonomic units,OTU),37.58%的OTU注释到门水平,24.36%的OTU注释到属水平;标准组阴道菌群Alpha多样性指数的Shannon指数、Chao1指数和ACE指数均显著低于肥胖组和偏瘦组(P<0.05);各组阴道菌群的Good’s coverage指数均在99.4%以上,表明样本测序质量较高;在门水平上,偏瘦组、标准组和肥胖组妊娠初期母牛阴道中前10大优势菌门为厚壁菌门(Firmicutes)、拟杆菌门(Bacteroidetes)、变形菌门(Proteobacteria)、无壁菌门(Tenericutes)、放线菌门(Actinobacteria)、梭杆菌门(Fusobacteria)、疣微菌门(Verrucomicrobia)、未知菌门(Unidentified_Bacteria)、螺旋体菌门(Spirochaetes)和Melainabacteria;在属水平上,偏瘦组、标准组和肥胖组前10大优势菌属分别为拟杆菌属(Bacteroides)、阿利斯佩斯菌属(Alistipes)、气球菌属(Aerococcus)、费氏菌属(Faecalibacterium)、链杆菌属(Streptobacillus)、链球菌属(Streptococcus)、棒杆菌属(Corynebacterium)、嗜组织菌属(Histophilus)、脲原体菌属(Ureaplasma)和支原体属(Mycoplasma),其中标准组嗜组织菌属显著高于其他2组(P<0.05)。

Comparison of the vaginal flora diversity of cows in early pregnancy with different body conditions

Abstract:
In order to investigate the effects of different body conditions on the vaginal flora of cows in early pregnancy, the vaginal secretions of 18 Simmental cows(6 cows in each group, fat cattle group、middle cattle group and thin cattle group)in the early pregnancy under the same grazing conditions were collected. The diversity of vaginal flora was analyzed by 16S rRNA sequencing. The results showed that: A total of 70 567 sequences were measured in average for each of the 18 samples, an average of 68 309 sequences were obtained, and the effective rate of quality control was 96. 84% after quality control; And 2 677 operational taxonomic units(OTU)were obtained through clustering, and among which 37. 58% OTU were annotated to phylum level and 24. 36% OTU were annotated to genus level; The Shannon index, Chao1 index and ACE index of Alpha diversity index of vaginal flora in MC group were significantly lower than those in the FC group and the TC group(P<0. 05). The Good's coverage index of vaginal flora in each group was more than 99. 4%, indicating that the sequencing quality of the samples was high. At the phylum level, the top 10 dominant phylum in the vagina of cows in the TC group, MC group and FC group in early pregnancy were Firmicutes, Bacteroidetes, Proteobacteria, Tenericutes, Actinobacteria, Fusobacteria, Verrucomicrobia, unidentified_Bacteria, Spirochaetes and Melainabacteria, respectively; At the genus level, the top 10 dominant genus in TC group, MC group and FC group were Bacteroides, Alistipes, Aerococcus, Faecalibacterium, Streptobacillus, Streptococcus, Corynebacterium, Histophilus, Ureaplasma and Mycoplasma, respectively. Among them, the Histophilus in the MC group was significantly higher than that in the other two groups(P<0. 05).
