原儿茶酸在鸡组织中的残留消除规律

摘要:
为研究原儿茶酸在鸡组织中残留消除规律,利用高效液相色谱法(HPLC)在260 nm处检测原儿茶酸在鸡组织中残留情况,确定流动相体积比为乙腈∶水=9∶91,用H3PO4调pH至2.5。结果表明:试验测得鸡肝、肾、肌肉、皮肤和脂肪中检测限分别为13.5、25.0、12.0和16.0 ng/g,定量限为45、85、40和50 ng/g;平均加标回收率为63.30%~102.65%,变异系数为1.01%~11.43%。以临床推荐剂量20 mg/kg(体重)口服给药,每天2次,连续5 d,肾脏、肝脏中原儿茶酸的残留量最高,皮肤和脂肪、肌肉中残留量较低。休药4 h后,各组织中原儿茶酸残留量均低于最高残留限量(肝脏1.52 mg/g,肾脏3.05 mg/g,肌肉0.51 mg/g,皮肤和脂肪3.05 mg/g)。试验结果表明,原儿茶酸在鸡组织中的残留消除较快,新药休药期的制定应根据具体情况进一步研究确定,目前初步建议休药期为0 h。

Residue depletion of protocatechuic acid in chicken tissues

Abstract:
The research objective was to study the tissue residues of protocatechuic acid (PCA) in chicken.A high-performance liquid chromatography (HPLC) method was used to detect and determine the PCAresidues at 260 nm in chicken tissues.The mobile phase of HPLC was composed of acetonitrile and water with the volume ration of 9∶91 and adjusted to pH 2.5 with H3PO4.The limits of detection (LOD)were 13.5,25.0,12.0,16.0 ng/g,and the limits of quantitation(LOQ) were 45,85,40 and 50 ng/g for liver,kidney,muscle,fat and skin,respectively.Average recoveries were ranged from 63.30% to 102.65%,and coefficients of variance were ranged from 1.01% to 11.43%.After the chickens were orally administered successively of 20 mg PCA per kg of body weight twice every day for 5 days,the residue levels were higher in liver and kidney and lower in muscle,fat and skin.Concentrations of PCA in tissues were all lower than the maximum residue level(MRL) at 4 h after final administration.The results indicated that there existeda rapid depletion of PCA in chicken tissues.Although we suggested the withdrawal period of PCA should be 0 hour,the withdrawal period of new medicine should be made based on the national specific situation.
