内蒙古绒山羊JAG1生物信息学及表达模式分析

摘要:
为分析绒山羊JAG1蛋白结构特征,及其在不同毛囊生长期中的表达模式,从阿拉善种羊场选择2周岁、体重相近的3只内蒙古绒山羊母羊,利用生物信息学方法比对绒山羊JAG1氨基酸序列与其他物种的相似性,分析JAG1蛋白质理化性质和结构特征;利用免疫组化和实时荧光定量PCR检测其在内蒙古绒山羊皮肤中的表达部位和表达量。结果表明,绒山羊JAG1氨基酸序列与绵羊相似性可达99.5%,JAG1蛋白信号肽位于第1~29个氨基酸序列,含有一个剪切位点和一个跨膜结构,具有61个丝氨酸、26个苏氨酸、19个酪氨酸磷酸化位点,63个O-糖基化位点,6个N-糖基化位点,属于不稳定亲水蛋白。在毛囊退行期(1月份)和休止期(4月份)未检测到JAG1蛋白的表达;生长前期(7月份)和生长期(9月份)在毛乳头细胞中表达。JAG1 mRNA在内蒙古绒山羊皮肤毛囊的4个时期均有表达,随着毛囊进入生长期JAG1 mRNA表达量逐渐增加,生长期皮肤中的表达量显著高于其他阶段(P<0.05)。综上,绒山羊JAG1氨基酸序列在物种进化过程中比较保守,JAG1 mRNA在内蒙古绒山羊毛囊生长前期和生长期表达升高,为深入研究绒山羊JAG1基因对毛囊发育的调控提供理论依据。

JAG1 bioinformatics and expression pattern analysis of Inner Mongolia Cashmere goat

Abstract:
In order to analyze the structural characteristics of JAG1 protein in Cashmere goats and its expression pattern in different hair follicle growth periods, three two-year-old Inner Mongolia cashmere goat ewes with similar body weight were selected from Alashan breeding farm. The similarity of the amino acid sequence of JAG1 between Cashmere goat and other species was compared by bioinformatics method, and the physicochemical properties and structural characteristics of JAG1 protein were analyzed. Immunohistochemistry and real-time fluorescence quantitative PCR were used to detect the expression sites and amounts of JAG1 in Inner Mongolia Cashmere goat skin. The results showed that the similarity of JAG1 amino acid sequence in Cashmere goat and sheep was 99. 5%. The signal peptide of JAG1 protein was located in the amino acid sequence of 1-29 containing a splice site and a transmembrane structure. There were 61 serine, 26 threonine and 19 tyrosine phosphorylation sites. 63 O-glycosylation sites, and 6 N-glycosylation sites. It was an unstable hydrophilic protein. The expression of JAG1 protein was not detected in hair follicle catagen(January)and telogen(April). However, JAG1 protein was expressed in the dermal papilla cells at the early stage of anagen(July)and anagen(September)hair follicle. JAG1 mRNA was expressed in the skin of Inner Mongolia cashmere goat in the four stages of hair follicle. With the development of hair follicles, the expression level of JAG1 mRNA gradually increased, and the expression level of JAG1 mRNA in the skin during the anagen was significantly higher than that in other stages(P<0. 05). In conclusion, the animo acid sequence of JAG1 in cashmere goat is relatively conservative in the process of species evolution, and the expression of JAG1 gene is increased in the early stage of anagen of hair follicle of Inner Mongolia Cashmere goat. This study provides a theoretical basis for further study on the regulation of hair follicle development by JAG1 gene in Inner Mongolia Cashmere goat hair follicle.
