EGCG对急性热应激肉鸡抗氧化能力的影响

摘要:
为研究表没食子儿茶素没食子酸酯(EGCG)对急性热应激肉鸡血清及器官抗氧化能力的影响,选取144羽10日龄淮南麻黄公鸡,随机分为3组:对照组(Ⅰ组)、急性热应激组(Ⅱ组)和急性热应激+EGCG组(Ⅲ组),每组6个重复,每个重复8羽。Ⅰ组和Ⅱ组饲喂基础饲粮,Ⅲ组饲喂在基础饲粮中添加400 mg/kg EGCG的试验饲粮。至94日龄时,Ⅰ组环境温度维持为(22±1)℃,Ⅱ组和Ⅲ组升温至(33±1)℃,持续12 h后,采集血清、脾脏、肝脏及空肠,用比色法检测抗氧化指标,并用实时荧光定量(qRT-PCR)法检测肝脏核因子E2相关因子2(Nrf2)信号通路相关基因表达水平。结果表明:1)与Ⅰ组相比,Ⅱ组肉鸡脾脏指数,血清和脾脏中总超氧化物歧化酶(T-SOD)和过氧化氢酶(CAT)活性,肝脏中T-SOD和谷胱甘肽过氧化物酶(GSH-Px)活性及Nrf2、NAD(P)H醌氧化还原酶1(NQO1)、CATGSH-Px mRNA表达量显著降低(P<0.05),而空肠中丙二醛(MDA)含量,肝脏中MDA和蛋白羰基(PC)含量及Kelch样环氧氯丙烷相关蛋白1(Keap1)mRNA表达量显著升高(P<0.05);2)与Ⅱ组相比,Ⅲ组肉鸡脾脏中CAT活性,肝脏中GSH-Px活性及CAT mRNA表达量显著升高(P<0.05),而空肠和肝脏中MDA含量显著降低(P<0.05)。综上,饲粮中添加EGCG可提高急性热应激肉鸡的抗氧化能力,进而缓解急性热应激导致的氧化损伤,其机制可能与EGCG激活Nrf2信号通路关键抗氧化基因表达有关,为EGCG应用于肉鸡生产奠定了理论基础。

Effects of EGCG on antioxidant capacity of acute heat-stressed broilers

Abstract:
The aim of the study was to investigate the effects of epigallocatechin gallate(EGCG)on antioxidant capacity in serum and organs of acute heat-stressed broilers. A total of 144 10-day-old male Huainan partridge chickens were randomly divided into 3 groups(control(group Ⅰ), acute heat stress(group Ⅱ)and acute heat stress+EGCG(group Ⅲ))with 6 replicates in each group and 8 birds in each replicate. The basal diet was fed in group Ⅰ and group Ⅱ, and the basal diet supplemented with 400 mg/kg EGCG was fed in group Ⅲ. At the age of 94 days, the ambient temperature of group I was maintained at(22±1)℃, and that of group Ⅱ and group Ⅲ was raised to(33±1)℃ for 12 hours. And then serum, spleen, liver and jejunum samples were collected. The antioxidant indexes were detected by colorimetry. The expression levels of Nrf2 signaling pathway-related genes were determined by qRT-PCR. The results showed as follows: 1)Compared with group I, the spleen index, total superoxide dismutase(T-SOD)and catalase(CAT)activities in serum and spleen, T-SOD and glutathione peroxidase(GSH-Px)activities and nuclear factor E2 related factor2(Nrf2), NAD(P)H quinone oxidoreductase 1(NQO1), CAT, GSH-Px mRNA expression levels in liver were significantly decreased(P<0. 05). The content of malondialdehyde(MDA)in the jejunum, the contents of MDA and PC and the mRNA expression level of Kelch-like epichlorohydrin-related protein 1(Keap1)in liver were significantly increased(P<0. 05). 2)Compared with group Ⅱ, CAT activity in spleen, GSH-Px activities and CAT mRNA expression levels in liver in group III were significantly increased(P<0. 05), while MDA contents in jejunum and liver were significantly decreased(P<0. 05). In summary, dietary EGCG supplementation can improve antioxidant capacity of acute heat-stressed broilers and then alleviate the oxidative damage caused by acute heat stress. The mechanism may be related to EGCG activating the expression of key antioxidant genes in the Nrf2 signaling pathway. This study provided theoretical foundation for the application of EGCG in broiler production.
