猪ARID5B基因多态性及其与脂肪沉积性状的相关性分析

摘要:
为探究猪ARID5B基因多态性及其与脂肪沉积性状的相关性,在DNA水平上对藏猪、大白猪ARID5B(AT-rich interaction domain 5B)基因5′侧翼区进行了单核苷酸多态性(Single nucleotide polymorphism,SNP)位点筛选;在mRNA水平上对两猪种的肝脏、背脂和背最长肌组织中的ARID5B基因表达量进行了RT-qPCR检测;并对两猪种的背膘厚和背最长肌组织中IMF(Intramuscular fat)含量进行测定。结果表明:在ARID5B基因5’侧翼区,存在T-2 216 C和In-2 029 Del缺失2个连锁突变位点,藏猪与大白猪呈极显著差异(P<0.01);在相同组织不同品种间,ARID5B基因在肝脏、背脂和背最长肌中mRNA相对表达量趋势完全一致,且藏猪ARID5B基因表达量均极显著低于大白猪(P<0.01);藏猪背膘厚和IMF含量均极显著高于大白猪(P<0.01);经DNA、mRNA水平、背膘厚和IMF含量相关性分析发现,ARID5B基因mRNA相对表达量与背膘厚和IMF含量呈显著负相关(P<0.05)。

Polymorphism of ARID5B gene and its correlation with fat deposition traits in pigs

Abstract:
To explore the polymorphism of ARID5B gene and its correlation with fat deposition in pigs, single nucleotide polymorphism(SNP)loci was screened for ARID5B gene 5′-UTR of Tibetan pigs and Large White pigs at the DNA level. The ARID5B gene was detected by RT-qPCR at mRNA level in liver, backfat and longissimus dorsi muscle tissues of the two pig breeds. The content of backfat and IMF(Intramuscular fat)in the longest muscle tissues of the two pig breeds were measured. The results showed that in the 5′-UTR of ARID5B gene, there were two linkage mutations of T-2 216 C and In-2 029 Del deletion, and the difference between Tibetan pigs and Large White pigs was extremely significant(P<0. 01). The relative mRNA expression of ARID5B gene in liver, backfat and longissimus dorsi muscle were identical in different breeds. The content in the Tibetan pigs were significantly lower than that in the Large White pigs(P<0. 01). The backfat thickness and IMF content of Tibetan pigs were significantly higher than that of Large White pigs(P<0. 01). The correlation analysis of DNA and mRNA levels with backfat and IMF levels showed that the relative mRNA expression of ARID5B gene was significantly negative with the backfat thickness and IMF content(P<0. 05).
