灌饮鸡脾转移因子对蛋雏鸡肠黏膜机械屏障发育的影响

摘要:
为研究不同剂量鸡脾转移因子(TF)对蛋雏鸡肠黏膜组织学屏障发育的影响,选取200只体重和生长情况基本一致的5日龄SPF蛋雏鸡,随机分为低剂量组、中剂量组、高剂量组和对照组,每组50只。给蛋雏鸡分别灌饮低(0.10 mL/只)、中(0.25 mL/只)、高(1.00 mL/只)剂量的TF,用药1周,停药1周,间歇给药,分别于12、26、40日龄取材肠道组织,石蜡切片,HE染色,比较不同TF剂量组肠道组织学结构的变化。结果表明使用不同剂量的TF后,与对照组相比:12日龄时TF应用组小肠绒毛高度均有所升高,隐窝深度降低,尤其二者比值变化显著:十二指肠中、高剂量组分别显著增加26.00%、26.96%,空肠高剂量组显著增加29.45%,回肠各剂量组均显著增加(19.74%~29.32%);26日龄,肠黏膜结构变化与12日龄相似,十二指肠中、高剂量组绒毛高度和隐窝深度比值显著增加9.46%、9.90%,空肠只有高剂量组显著增加12.99%;回肠各剂量组显著增加(17.07%~25.60%);40日龄,十二指肠中、高剂量组比值分别显著增加17.97%、22.19%;空肠高剂量组显著增加28.68%;回肠各剂量组均显著增加(17.04%~25.56%)。因此,给蛋雏鸡灌饮高剂量TF可显著增加小肠绒毛高度和隐窝深度比值,且与应用剂量呈正相关,能够有效改善小肠的消化吸收功能。

Effect of drinking spleen transfer factor on development of intestinal mucosal barrier in layer chickens

Abstract:
In order to study the effects of different doses of chicken spleen transfer factor (TF) on the development of intestinal mucosal barrier in layer chickens,200 5-day-old SPF egg chicks with the same body weight and growth were selected and randomly divided into low-dose group,medium-dose group,high-dose group and control group with 50 chicks in each group.Chicks were treated with different doses of oral TF (0.10, 0.25 and 1.00 mL each) from 5 days old,in an intermittent administration way with one week of drug administration and one week of drug withdrawal.Intestinal tissues were collected at 12, 26 and 40 days old.Paraffin section and HE staining was performed.Histological changes were compared in different TF dose groups.Compared to that of control group,the results showed that at 12 days of age,the height of intestinal villi increased and the depth of crypt decreased,especially the ratio of the two changed significantly:ratios of duodenum with the middle and high dose groups were significantly increased 26.00% and 26.96% compared with the control group;In jejunum,the ratio was obviously increased 29.45% in high dose group;it increased significantly (19.74%-29.32%) in ileum each group.At 26 days of age,the changes of intestinal mucosal structure were similar to those of 12 days old,especially,the ratio of villus height and crypt depth of duodenums was increased significantly in middle (9.46%) and high (9.90%) doses groups;there was a significant increase by 12.99% in jejunum in the high dose group;it increased significantly (17.07%-25.60%) in ileum each dose group.At 40 days of age,the ratio of duodenum increased significantly by 17.97% (middle doses) and 22.19% (high doses),respectively.The ratio of jejunum increased significantly by 28.68% in the high dose,and it increased significantly (17.04%-25.56%) in ileum each group.Therefore,the ratio of villus height and crypt depth of the small intestine was increased in the high dose TF feeding groups,and it was positively correlated with the applied dose.Therefore,TF can effectively improve the digestion and absorption function of the small intestine.
