不同pH和SCFAs对奶牛瘤胃上皮细胞SCFAs转运蛋白和GPR41表达的影响

摘要:
为研究不同pH和短链脂肪酸(SCFAs)对奶牛瘤胃上皮细胞SCFAs转运蛋白和G-蛋白偶联受体41(GPR41)表达影响,设计6组试验,每组3个重复,培养24 h之后,收集细胞提取细胞总RNA。结果表明:在添加SCFAs条件下,pH 6.2、6.8和7.4之间的MCT1表达量差异不显著(P > 0.05);pH 6.8且添加SCFAs条件下,MCT1的表达量显著高于缺乏SCFAs pH 6.8和7.4试验组(P<0.05);pH 6.2的MCT4的表达量显著高于pH 6.8和7.4(P<0.01),MCT4的表达量显著高于缺乏SCFAs(P<0.01),NHE1、NHE2和NHE3表达量显著高于缺乏SCFAs时的表达量(P<0.01);随着pH的降低,NHE1、NHE2和NHE3的表达量逐渐上调,而在缺乏SCFAs条件下,GPR41不表达;在添加SCFAs之后,显著激活了GPR41的表达。研究发现SCFAs能够显著加强奶牛瘤胃上皮细胞SCFAs转运蛋白和GPR41的表达量,从而证明细胞对SCFAs的吸收主要依赖于SCFAs转运蛋白和GPR41。

Effects of different pH and SCFAs on the expressions of SCFAs transport proteins and GPR41 in the rumen epithelial cells of dairy cow

Abstract:
To investigate the effects of different pH and SCFAs on the expression of SCFAs transport proteins and GPR41 in rumen epithelial cells of dairy cows. Six groups of experiment were designed, after 24 h culturing, cells were collected and RNA was extracted. The results showed that with SCFAs supplementation:There was no significant difference in the expression levels of MCT1 among pH 6.2, 6.8 and 7.4 (P>0.05). The expression of MCT1 in pH 6.8 was significantly higher than that in pH 6.8 and 7.4 (P<0.05); The expression of MCT4 in pH 6.2 was significantly higher than those in pH 6.8 and pH 7.4 (P<0.01); The expression of MCT4 was significantly higher than those of SCFAs (P<0.01). The expressions of NHE1, NHE2 and NHE3 were significantly higher than those without SCFAs supplement (P<0.01). With the decreasing of pH, the expressions of NHE1, NHE2 and NHE3 increased gradually; Without additional SCFAs, GPR41 was not expressed. After the SCFAs supplementation, the expression of GPR41 was greatly activated. In conclusion, SCFAs could significantly enhance the expressions of SCFAs transport proteins and GPR41 in rumen epithelial cells of dairy cow. It was proved that the cell uptake of SCFAs was mainly depended on SCFAs transporter and GPR41.
