黄瓜YABBYs家族及CsYAB1a基因功能初探

摘要:
为鉴定黄瓜YABBYs家族及对CsYAB1a基因的生物学功能进行初步探究,通过BLASTp比对鉴定黄瓜YABBYs基因,并对其进行基因、蛋白质水平分析和染色体定位,同时利用实时荧光定量和原位杂交技术检测其在黄瓜中的时空表达模式;在拟南芥中异源过表达CsYAB1a基因,对转基因株系进行表达量分析和表型观察统计;通过外源喷施脱落酸和生长素处理检测CsYAB1a基因的响应情况。结果表明:1)黄瓜YABBYs家族包含8个家族成员,所有成员均包含C2C2锌指结构域和YABBY结构域;大部分YABBYs基因在生殖器官有表达,CsYAB1a基因主要在花瓣原基和子房外果皮中富集,并呈现出远轴端表达的极性模式。2)在拟南芥中异源过表达CsYAB1a基因导致雌雄蕊发育畸形且果荚变短、叶片细长并向远轴端卷曲等表型;CsYAB1a基因调控拟南芥果实和叶片的发育,并对果实发育基因AtSPL、AtINDAtHEC以及叶片极性基因AtAS1AtKANs的表达量有影响。3)黄瓜YABBYs基因启动子包含5种激素响应元件,CsYAB1aCsCRC基因对外源脱落酸和生长素喷施处理均有响应。综上,推测黄瓜YABBYs家族可能在黄瓜营养生长和生殖生长过程中发挥重要作用,且CsYAB1a基因调控黄瓜果实和叶片的发育。

Preliminary study on the function of YABBYs family and CsYAB1a gene in cucumber(Cucumis sativus L.)

Abstract:
To identify CsYABBYs family in cucumber and explore the biological function of CsYAB1a gene preliminarily, CsYABBYs genes were identified by BLASTp homology comparison. Analysis on gene and protein sequence and chromosomal localization also was performed, then their spatio-temporal expression patterns were detected by RT-qPCR and in situ hybridization. CsYAB1a gene was overexpressed in Arabidopsis, and transgenic lines' expression level and phenotype were analyzed. The expression level of CsYAB1a gene was measured after treated with different kinds of hormones. The results were as follows: 1) CsYABBYs family consists of 8 members, all of which contain the C2C2 zinc finger and YABBY domain; Most of the YABBYs genes were expressed in reproductive organs, CsYAB1a gene was mainly enriched in petal primordium and ovary exocarp and showed the polar pattern of distal expression. 2)In Arabidopsis, heterologous overexpression of CsYAB1a gene resulted in phenotypes including malformed pistils and stamens, shorter siliques and elongated distal curling leaves; CsYAB1a gene regulates the development of fruits and leaves and affects the expression levels of fruit development genes AtSPL, AtIND, and AtHEC and leaf polarity genes AtAS1 and AtKANs. 3)The promoter of the CsYABBYs gens contains five hormone response elements, and both CsYAB1a and CsCRC genes responded to exogenous abscisic acid and auxin spraying treatments. In conclusion, CsYABBYs family might play essential roles in the vegetative and reproductive growth of cucumbers, and CsYAB1a gene may involve fruit and leaf development.
