生态隔间影响韭菜内生细菌、根际和叶围细菌的定殖及互作

摘要:
为阐明3种不同品种韭菜的内生细菌、叶围和根际5个生态隔间中细菌群落组成的变化及各生态隔间相关网络的结构差异,通过16S rRNA基因测序及分析,对不同功能的微生物群落多样性进行了讨论,分析了功能微生物在各个生态隔间中的丰度变化及地上隔间、地下隔间和各品种的微生物共现网络。结果表明:1)‘久星16号’韭菜品种叶围微生物中与病原菌防御相关的假单胞菌属(Pseudomonas)含量显著高于其他品种;2)生态隔间对于门和属水平微生物群落组成的相对丰度具有显著性影响,鞘氨醇单胞菌属(Sphingomonas)在植物内部呈现由上至下逐渐递减的趋势;3)地下生态隔间的微生物共现网络较地上隔间复杂性和抗干扰性更强,韭菜各品种间存在网络直径相似但中心节点不同的微生物共现网络,‘久星16号’‘久星18号’因其应对温度变化特性,在变形菌门(Proteobacteria)、酸杆菌门(Acidobacteria)与绿弯菌门(Chloroflexi)间拥有更多负相关作用。综上,生态隔间对韭菜微生物的结构分布和互作网络的影响要高于韭菜品种,且不同韭菜品种会根据自身特性招募特异微生物。

Effects of ecological compartments on the composition and co-occurrence network of endophytic bacteria, rhizosphere and phylloplane bacteria in Chinese chives

Abstract:
This study aimed to clarify the changes of bacterial community composition and the structural differences of related networks in three different varieties of Chinese chives endophytes and five ecological compartments of phylloplane and rhizosphere. Based on 16S rRNA gene sequencing and analysis. The diversity of microbial communities with different functions were discussed. The abundance of functional microorganisms in each ecological compartment and the microbial co-occurrence network of aboveground compartment, underground compartment and various varieties were analyzed. The results showed that: 1)The content of Pseudomonas related to pathogen defense in phylloplane of ‘Jiuxing 16' leek variety resistant to gray mold was significantly higher than that of other varieties; 2)Ecological compartments had significant effects on the relative abundance of microbial community composition at phylum level and genus level. Sphingomonas gradually decreased from top to bottom in plants; 3)The microbial co-occurrence network of underground ecological compartment was more complex and anti-interference than that of aboveground compartment. There were microbial co-occurrence networks with similar network diameters but different central nodes among Chinese chives varieties. There were more negative connections between Proteobacteria, Acidobacteria and Chloroflexi, because ‘Jiuxing 16' and ‘Jiuxing 18' displayed different dormancy characteristics. In summary, the effect of ecological compartment on the microbial structure distribution and interaction network of leek was higher than that of leek varieties, and different leek varieties would recruit specific microorganisms according to their own characteristics.
