豫北夏玉米根际2种根腐线虫的形态和分子鉴定

摘要:
为明确采集于豫北地区鹤壁市和焦作市玉米根际土壤样品中根腐线虫的种类,采用改良的贝尔曼漏斗法分离土壤样品中的根腐线虫,通过光学显微镜进行形态鉴定,并基于rDNA 28S D2-D3区和rDNA-ITS区的序列对采集的根腐线虫进行分子鉴定。形态学与分子鉴定的结果均表明:来源于鹤壁市的HB-1种群为斯克里布纳短体线虫(Pratylenchus scribneri),来源于焦作市的JZ-257种群为落选短体线虫(P. neglectus)。基于rDNA 28S D2-D3区和rDNA-ITS区序列构建的系统进化树结果表明:供试2种短体线虫分别与GenBank数据库中其它斯克里布纳短体线虫和落选短体线虫处于同一高度支持的分支。本研究结果为玉米根腐线虫病害的识别与防控提供了科学依据。

Morphological and molecular identification of two root-lesion nematodes in the rhizosphere of summer maize in northern Henan

Abstract:
In order to identify the species of root-lesion nematodes in maize rhizosphere soil samples collected in Hebi and Jiaozuo in Northern Henan, the modified Baermann funnel method was adopted to isolate the root-lesion nematodes from the collected soil samples, morphological identification was performed by optical microscope, and based on the sequence of rDNA 28S D2-D3 region and rDNA-ITS region, the collected root-lesion nematodes were identified. The results of both morphological and molecular identification showed that HB-1 population from Hebi City was P. scribneri, and JZ-257 population from Jiaozuo City was P. neglectus. The phylogenetic tree constructed based on the sequences of rDNA 28S D2-D3 region and rDNA-ITS region showed that the two species were in the same highly supported clads as other P. scribneri and P. neglectus in GenBank database, respectively. The results of this study provided a scientific basis for the identification and control of corn root-lesion nematodes disease.
