一株犬副流感病毒CPIV-BJ01株全基因组测序与其遗传变异分析

摘要:
为分析本实验室分离的犬副流感病毒CPIV-BJ01株全基因组序列与其遗传变异情况,对CPIV-BJ01株全长进行分段扩增并测序,拼接获得全基因组序列,与GenBank发布的27株副流感病毒5型分别进行全基因组和包膜糖蛋白(F、SH和HN)核苷酸及氨基酸序列的相似性比对,分析其遗传进化关系。结果显示,CPIV-BJ01毒株与犬源PIV5 1 168-1亲缘关系最近,核苷酸相似性为99.9%,氨基酸相似性为99.8%。CPIV-BJ01的FHN基因序列变化较为保守,核苷酸和氨基酸变化均在4%以内,其中F蛋白的氨基酸变化形成一处新的O-糖基化位点;CPIV-BJ01株的SH基因序列与其他演化分支毒株差异显著,核苷酸相似性为84.4%~97.0%,氨基酸相似性为31.8%~95.6%。综上,CPIV-BJ01株基因组序列与前人报道的毒株相比,在F、HN基因和部分非编码区存在核苷酸或氨基酸序列的改变,同时F蛋白存在糖基化修饰的改变,均为该毒株所特有,该研究结果丰富了中国CPIV流行株的基因组信息。

Complete genome sequencing and genetic variation analysis of an isolated canine parainfluenza virus strain CPIV-BJ01

Abstract:
To analyze the complete genome sequence and genetic variation of canine parainfluenza virus strain CPIV-BJ01 isolated in laboratory, the full length of CPIV-BJ01 strain was amplified and sequenced. The nucleotide and amino acid of the whole genome and envelope glycoproteins(F, SH and HN)of 27 parainfluenza virus type 5 strains from GenBank database were compared. The genetic and evolutionary relationships were analyzed. The results showed that CPIV-BJ01 strain had the closest genetic relationship with canine PIV5 1 168-1, with nucleotide similarity of 99. 9% and amino acid similarity of 99. 8%. The F and HN gene sequences of CPIV-BJ01 were relatively conservative. Their changes of nucleotide and amino acid were within 4%, among which the amino acid of F protein formed a new O-glycosylation site. The SH gene sequence of CPIV-BJ01 was significantly different from that of other evolutionary branches with nucleotide similarity of 84. 4%-97. 0% and amino acid similarity of 31. 8%-95. 6%. In summary, compared with the previously reported strains, the genomic sequence of CPIV-BJ01 strain had nucleotide or amino acid sequence changes in F, HN gene and some non-coding genes, and there are glycosylation modifications in F protein, which were unique of the CPIV-BJ01 strain. These results enriched the genome information of Chinese CPIV epidemic strains.
