春小麦脂肪氧化酶TaLox-B位点等位变异检测及分布规律

摘要:
为阐明脂肪氧化酶TaLox-B位点等位基因在国内外不同地区春小麦种质资源中的分布规律,利用已开发的功能基因标记LOX16、LOX18和LOX-B23对154份国内新疆春小麦地方品种(系)和83份国外春小麦品种(系)进行分子标记检测。结果表明,237份春小麦品种(系)在TaLox-B1、TaLox-B2和TaLox-B3位点上与高LOX活性相关的等位基因TaLox-B1aTaLox-B2aTaLox-B3a的分布频率分别为:8.0%、93.2%和86.1%,与低LOX活性相关的等位基因TaLox-B1bTaLox-B2bTaLox-B3b的分布频率分别为:92.0%、6.8%和13.9%。新疆维吾尔自治区四大生态区的春小麦地方品种(系)中,东疆地区和伊犁河谷地区的参试材料未发现优异等位变异TaLox-B1a;而优异等位变异TaLox-B2aTaLox-B3a的分布频率由高到低为:东疆地区>北疆地区>南疆地区>伊犁河谷地区。供试品种(系)的国外不同地区春小麦品种(系)中,优异等位变异TaLox-B1a的分布频率由高到低为:欧洲地区>非洲地区>美洲地区>亚洲地区。本研究所用春小麦品种(系)共检测出5种基因型组合:TaLox-B1a/TaLox-B2a/TaLox-B3aTaLox-B1b/TaLox-B2a/TaLox-B3aTaLox-B1a/TaLox-B2a/TaLox-B3bTaLox-B1b/TaLox-B2a/TaLox-B3bTaLox-B1b/TaLox-B2b/TaLox-B3b,分别与高、中和低LOX活性相关,其频率分别为86.1%、7.2%和6.7%。新疆四大生态区的春小麦地方品种(系)中,东疆地区材料均检测为具有最高LOX活性基因组合TaLox-B1b/TaLox-B2a/TaLox-B3a。供试的国外春小麦品种(系)中,美洲地区的材料具有高LOX活性基因型组合TaLox-B1a/TaLox-B2a/TaLox-B3aTaLox-B1b/TaLox-B2a/TaLox-B3a的频率分别为13.8%和69.0%,总体高于国外其他地区。

Molecular identification and distirbution of lipoxygenase TaLox-B loci in spring wheat

Abstract:
In order to clarify the distribution of allele of TaLox-B locus in spring wheat germplasm resources in different regions at domestic and abroad, this study used the developed functional gene markers LOX16, LOX18 and LOX-B23 to detect the molecular markers of 154 domestic Xinjiang spring wheat local varieties and 83 abroad spring wheat varieties. The results showed that: The distribution frequencies of alleles TaLox-B1a, TaLox-B2a and TaLox-B3a related to high LOX activity at TaLox-B1, TaLox-B2 and TaLox-B3 loci in 237 spring wheat varieties were 8. 0%, 93. 2% and 86. 1%, and the distribution frequencies of the alleles TaLox-B1b, TaLox-B2b and TaLox-B3b associated with low LOX activity were 92. 0%, 6. 8% and 13. 9%. Among the local varieties of spring wheat in the four major ecological zones in Xinjiang Uygur Autonomous Region, no excellent allelic variation TaLox-B1a was found in the tested materials in eastern Xinjiang and Yili River Valley. However, the distribution frequency of excellent allelic TaLox-B2a and TaLox-B3a were as follows: Eastern Xinjiang>Northern Xinjiang>Southern Xinjiang>Yili River Valley. Among the tested varieties of spring wheat varieties in different regions abroad, the distribution frequency of excellent allelic variation TaLox-B1a was as follows: European>African>American>Asian. The analysis of 3 loci genotype combinations indicated that there are 5 kinds of genotypes combinations existed in the spring wheat varieties used in this study: TaLox-B1a/TaLox-B2a/TaLox-B3a or TaLox-B1b/TaLox-B2a/TaLox-B3a, TaLox-B1a/TaLox-B2a/TaLox-B3b or TaLox-B1b/TaLox-B2a/TaLox-B3b and TaLox-B1b/TaLox-B2b/TaLox-B3b. They were associated with the high, medium and low LOX activity, and the distribution frequencies were 86. 1%、7. 2% and 6. 7%, respectively. Among the local varieties of spring wheat in the four major ecological regions of Xinjiang, the materials in eastern Xinjiang were all detected as having the high LOX active gene combination TaLox-B1b/TaLox-B2a/TaLox-B3a. Among the tested varieties of spring wheat varieties in different regions abroad. The tested materials in the Americas had the high LOX activity gene combinations TaLox-B1a/TaLox-B2a/TaLox-B3a or TaLox-B1b/TaLox-B2a/TaLox-B3a were 13. 8% and 69. 0%, respectively, which higher than other regions abroad.
