蝴蝶兰(Phalaenopsis spp.)与钻喙兰(Rhynchostylis retusa)远缘杂交种子无菌播种及后代表型分析

摘要:
选择蝴蝶兰(Phalaenopsis spp.)与钻喙兰(Rhynchostylis retusa)进行属间正反远缘杂交,杂交组合共计36 个,获得远缘杂种后代141 株。采用无菌播种技术和性状调查的方法分别对种子和杂种后代进行分析,研究结果表明:1)36 个杂交组合共杂交180 朵花,有12 个组合座果,共获20 个果荚,座果率为11.1%;2)在座果的12 个组合中,仅有1 个组合PR01的种子在诱导培养基上萌发,而其他11个组合的果荚可能由于发育不充分、种胚未能形成,导致种子不能萌发;3)PR01的种子在诱导培养基M1、M2、M3等3 种培养基上均能萌发,在增殖过程中,BA 2.0~3.0 mg/L及Ad 2.0~3.0 mg/L有利于PR01原球茎的增殖,同时生根培养基T2适宜PR01壮苗生根;4)PR01远缘杂种F1无论在数量性状上,还是在假质量性状上大都介于双亲之间,遗传了双亲的特性,但开花期不一致,其种植3 年后其开花率为15.6%,而亲本全部开花且整齐一致。PR01植株及开花性状总体上趋于蝴蝶兰。

Aseptic seeding and F1 phenotypic analysis of intergeneric hybrid between Phalaenopsis spp. and Rhynchostylis retusa

Abstract:
Phalaenopsis spp. and Rhynchostylis retusa were selected for intergeneric hybridization study. A total of 141 progenies were obtained from 36 cross combinations. The sterile sowing technique and character investigation were used to analyze the seeds and hybrid progenies. The results showed that: 1)Among 36 cross combinations, only 12 cross combinations obtained 20 capsules and the fruit-setting rate was 11. 1%. 2)Of the 12 combinations with the fruit-settings, only one combination PR01 seeds germinated on the induction medium, and the rest of 11 combinations could not germinate, which could be due to insufficient development of capuls and failure during embryo formation. 3)The seeds of PR01 could germinate on the induction medium of M1, M2 and M3. BA 2. 0-3. 0 mg/L and Ad 2. 0-3. 0 mg/L were beneficial to protocorm proliferation and the rooting medium T2 was suitable for the rooting of PR01. 4)The quantity and pseudo-quality traits of the hybrid F1 traits displayed were mostly somewhere in between both parents. The hybrid F1 generation inherited the characteristics of both parents. However, their flowering time was inconsistent and the flowering rate was 15. 6% after 3 years of planting, whereas both parents all blossomed at the same time. In conclusion, the plants and flowering traits of PR01 generally tend to be Phalaenopsis.
