海岛棉转录因子基因GbTCP44的克隆及特征分析

摘要:
为阐明TCP基因在棉花纤维发育中的功能,以海岛棉‘新海21号’为材料,运用RT-PCR技术克隆得到海岛棉GbTCP44基因的cDNA序列,并借助生物信息学、亚细胞定位、实时荧光定量PCR技术分析预测其结构和功能。结果表明:1)GbTCP44基因的编码区序列全长为1 035 bp,共编码344个氨基酸,预测分子式为 C1618H2538N484O525S13,分子量约为37.59 ku,等电点为8.83,其序列(N端)有一个高度保守的TCP功能结构域,属于TCP转录因子家族Class I成员;2)GbTCP44基因在细胞核中表达,属于核表达蛋白,该蛋白的结构域与其他植物TCP蛋白具有高度的保守性,并与雷蒙德氏棉的亲缘关系最近;3)GbTCP44基因在花器官中的表达量显著高于营养器官的表达量;该基因在棉纤维发育20 d时的表达量均为最高;当棉纤维发育至25和30 d时,该基因在长纤维品种中的表达量低于在短纤维品种中的表达量,且存在显著差异(P<0.05),表明GbTCP44基因可能参与棉花纤维伸长期的纤维细胞伸长的发育。

Cloning and characterization of a transcription factor gene GbTCP44 in Gossypium barbadensse

Abstract:
To better understand the function of cotton TCP genes in cotton fiber development, Gossypium barbadensse ‘Xinhai 21' was taken as study material, and cDNA sequence of GbTCP44 was obtained through RT-PCR technology. The structure and function of GbTCP44 were predicted by using bioinformatics methods, subcellular localization, and qRT-PCR technology. The results showed that: 1)The GbTCP44 gene has 1 035 bp open reading frame, which encodes 344 amino acids. Its molecular formula is C1618H2538N484O525S13 with a predicted molecular weight of approximately 37. 59 ku and a isoelectric point of 8. 83. The N-terminus sequence has a typical TCP conservative domain, which belongs to the Class I member of the TCP transcription factor family; 2)The GbTCP44 gene is a nuclear-expressing protein, which is expressed in the nucleus. The protein domain is highly conserved with other plant TCP proteins and has the closest relationship with Gossypium raimondii. 3)The expression of GbTCP44 gene in flower organs is significantly higher than that in vegetative organs; The expression level of this gene is the highest in cotton fibers at 20 d; When cotton fiber development reached 25 and 30 d, the expression level of the gene in long-fiber varieties is lower than that in short-fiber varieties, and there was a significant difference(P<0. 05), In conclusion, GbTCP44 gene might be involved in the development of fiber cell elongation during cotton fiber elongation.
