稀土壳糖胺螯合盐对热应激黄羽肉鸡脂肪组织学和ABCG1表达的影响

摘要:
为探讨稀土壳糖胺螯合盐(RECC)对热应激黄羽肉鸡脂肪组织学及三磷酸结合盒转运体G1(ATP-binding cassette G1,ABCG1)mRNA表达的影响。随机选取90只饲养至29日龄黄羽肉鸡,分为3个处理组,即常温对照组、热应激组和RECC组(热应激+基础日粮中添加0.03% RECC)。分别在热应激处理1 和2 周后,采集肉鸡内脏、皮下及肌间脂肪,实时荧光定量(qRT-PCR)方法检测脂肪细胞大小变化及脂肪组织中三磷酸结合盒转运体G1 mRNA表达变化。结果表明:热应激1 周后,与对照组和热应激组相比,RECC组肉鸡内脏、皮下和肌间脂肪细胞直径与相比显著减小(P<0.01),3 种脂肪组织中ABCG1 mRNA表达水平显著升高(P<0.05);热应激 2周后,与对照组和热应激组相比,RECC组肉鸡内脏和肌间脂肪细胞直径显著减小(P<0.05),皮下脂肪细胞直径无显著变化。ABCG1 mRNA表达水平的检测结果表明:与热应激组相比,RECC组内脏和皮下脂肪组织中ABCG1 mRNA表达水平显著降低(P<0.01)。综上所述,RECC可通过减小脂肪细胞直径和促进ABCG1的表达,从而抑制热应激1周肉鸡体内脂肪沉积。

Effects of rare earth chitosan chelate on adipose histology and ABCG1 mRNA expression in thermal stressed broilers

Abstract:
The aim of this study was to investigate the effects of rare earth chitosan chelate salt(RECC)on adipose histology and the expression levels of ATP-binding cassette G1(ABCG1)mRNA in yellow feather broilers treated with thermal stress. A total of 90 29-day-old yellow-feathered broilers were randomly divided into 3 treatment groups: Normal temperature control group, thermal stress group and RECC group(thermal stress + basal diet added 0. 03% RECC). The visceral, subcutaneous and intermuscular fats of broilers were collected after 1 and 2 weeks of thermal stress treatment. The size of adipocytes and the expression levels of ABCG1 mRNA in each adipose tissue were determined by qRT-PCR. The results showed that the diameter of adipocytes in visceral, subcutaneous and intermuscular was significantly reduced in the RECC group compared with the control group and the thermal stress group(P<0. 01)after 1 week treated with thermal stress, while the expression level of ABCG1 mRNA was significantly increased(P<0. 05). The diameter of adipocytes in visceral and intermuscular in RECC group was significantly decreased compared with control group and thermal stress group(P<0. 05)after 2 weeks treated with thermal stress; The expression levels of ABCG1 mRNA in visceral and subcutaneous adipose tissues in RECC group were significantly decreased compared with thermal stress group(P<0. 01). In summary, RECC could inhibit the fat deposition through decreasing the diameters of adipocytes and increasing the level of ABCG1 mRNA in broilers treated with thermal stress for 1 week.
