基于SPR生物传感器的H5N1流感病毒快速检测方法

摘要:
为利用表面等离子共振(Surface Plasmon Resonance,SPR)生物传感器建立一种快速检测H5N1流感病毒的新方法。该研究利用SPR生物传感器以及H5N1单克隆抗体,对H5N1流感病毒样品进行检测分析。结果表明:该方法特性如下:1)可实现针对H5N1流感病毒的直接、快速和实时检测,整个过程无需任何标记;2)检测限为104.44 TCID50/mL,低于ELISA方法的103.24 TCID50/mL;3)在抗体消耗量方面,SPR技术要优于ELISA技术近100倍。

Rapid and sensitive detection of H5N1 virus based on surface plasmon resonance (SPR)

Abstract:
An efficient alternative method to detect the presence of H5N1 virus based on surface plasmon resonance (SPR) and monoclonal antibody against Influenza A (H5N1) virus was proposed in this study. Samples of H5N1 influenza virus were tested and analyzed. The results showed:1) The good performance of SPR biosensor was based on the direct detection, real time record, and without any additional labeling. 2) The detection limit of the assay was found to be 104.44 TCID50/mL, which was lower than the limit of ELISA of 103.24 TCID50/mL. 3) The consumption of antibody in SPR test was 100-fold lower than that in ELISA test. Thus, in the future, with the development of high quality virus antigen and antibody production, the SPR biosensor system would become the most commonly applied and efficient approach for influenza A viruses diagnosis.
