长双歧杆菌L-DT的高密度培养

摘要:
针对益生菌株长双歧杆菌L-DT (Bifidobacterium longum ssp.longum L-DT)产品线单一,菌株应用需多元化的问题,采用高密度培养技术,利用食品级原料,通过单因素试验、Plackett-Burman试验、最陡爬坡试验和响应面分析优化液态培养基和培养条件。结果表明:适合L-DT生长的培养基中各成分的质量分数为,葡萄糖1.5%、乳糖2.4%、酵母浸粉2.26%、酵母蛋白胨2%、低聚异麦芽糖1.5%、西红柿汁3%;培养条件为,菌液接种量4.46%、初始pH6、温度37℃。培养得到的菌液OD600为1.113,与模型预测值1.104无显著性差异,L-DT活菌值达1.5×109 CFU/mL。在7 L发酵罐中进行放大培养验证,L-DT活菌数最高可达5×109 CFU/mL,是试管培养的3.33倍。

High density culture of Bifidobacterium longum L-DT

Abstract:
In view of the single product line of probiotic strain L-DT (Bifidobacterium longum ssp. longum L-DT), and its application to be diversified, a high density culture technology with food grade raw materials was used to optimize the liquid medium and culture conditions. Single factor experiment, Plackett-Burman experiment, steepest climbing experiment and response surface analysis were adopted in this study. The results showed that:The mass fraction of each component in the medium suitable for L-DT growth was:glucose 1.5%, yeast peptone 2%, oligomeric maltose 1.5%, tomato juice 3%, lactose 2.4% and yeast extract 2.26%. The culture condition was:Inoculation quantity (volume fraction) 4.46%, pH 6 and temperature at 37℃. Under this condition, the OD600 of the bacterial liquid could reach 1.113, which had no significant difference compared to the predicted value of 1.104. And the quantity of viable bacteria could be up to 1.5×109 CFU/mL. Amplification and verification in a 7 L fermenter, the maximum number of L-DT viable bacteria reached 5×109 CFU/mL, which was 3.33 times higher than that in tube culture.
