姜黄素及细菌素对子宫内膜炎大鼠血清中抗炎活性的影响

摘要:
为研究姜黄素(Curcumin)及细菌素(nisin)对患大鼠子宫内膜炎血清中基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶-1(TIMP-1)、环氧合酶-2(COX-2)、肿瘤坏死因子α (TNF-α) 的作用,构建大鼠子宫内膜炎模型,将160只SD大鼠随机分为细菌素组、姜黄素组、阳性对照组及生理盐水组,建模型前0 h和试验后24 、48 、72 h,用ELISA试剂盒检测各试验组中MMP-9、基质金属蛋白抑制物-1(TIMP-1)、环氧合酶-2(COX-2)及TNF-α的变化情况来监测治疗效果。结果表明:1)在0~72 h之间,建模0 h前时,细菌素组和姜黄素组中MMP-9的浓度显著高于生理盐水组和阳性对照组(P<0.05),生理盐水组中MMP-9的浓度比其他各用药组显著降低(P<0.05);在各药物组的作用下,MMP-9浓度的升高常伴随着TIMP-1的浓度的升高;2)各用药组均能降低血清中TNF-α的浓度,而生理盐水组0~72 h时间点时血清中TNF-α的浓度持续升高,血清中TNF-α的浓度随着COX-2的浓度的升高而降低。由此可见,细菌素和姜黄素能显著提高子宫内膜炎大鼠血清中TIMP-1和COX-2的表达,抑制MMP-9和TNF-α的活性而发挥调控子宫内环境动态平衡,表明细菌素和姜黄素对MMP-9、TIMP-1、COX-2的浓度具有正调控作用,而对TNF-α恰好相反。

Influence of curcumin and nisin on the anti-inflammatory activity in the serum of endometritis rats

Abstract:
In order to study the influence of curcumin and nisin on matrix metalloproteinase-9 (MMP-9),matrix metalloproteinase (TIMP-1) and cyclooxygenase-2 (COX-2),tumor necrosis factor α (TNF-α ) function in the endometritis of rats serum,the endometriosis model of rats was established.A total of 160 SD rats were randomly divided into bacteriocin group,curcumin group,positive control group,and normal saline group.Samples were taken at 0 h before the establishment of the model and 24,28,and 72 h after the test,and ELISA kit was used to detect MMP-9 in each experimental group,(TIMP-1),cyclooxygenase-2 (COX-2) and TNF-α changes was used to monitor the effects of treatment.The results showed that:1) Before modeling,establishment of the model,the concentrations of MMP-9 in bacteriocin group and curcumin group were significantly higher than that of saline group and positive control group (P<0.05);The concentration of MMP-9 in saline group was significantly lower than other groups (P<0.05);Under the influence of the curcumin and nisin groups,MMP-9 concentration was often associated with the increasing concentration of TIMP-1.2) The concentration of TNF-α in serum was reduced in curcumin and nisin groups.And the concentration of saline group between 0 h and 72 h increased,and serum TNF-α concentration decreased with increasing COX-2 concentration.The results showed that nisin and curcumin significantly increased TIMP-1 in the serum of rats with endometriosis and the expression of COX-2,inhibited the activity of MMP-9 and TNF-α and regulatory pathways and environmental balance inside the uterus.In conclusion,nisin and curcumin displayed positive effect on the concentrations of MMP-9,TIMP-1,COX-2,and negative effect on that of TNF-α.
