猪源支气管败血波氏杆菌的分离鉴定及其皮肤坏死素(DNT)粗提液的小鼠皮肤毒性试验

摘要:
为更好地进行波氏菌病的防治,利用分离培养、生化实验、PCR鉴定和同源性分析对采自四川某规模化猪场有明显萎缩性鼻炎症状的猪鼻腔拭子进行支气管败血波氏杆菌的分离鉴定,参考经典的皮肤坏死素(DNT)粗提技术,将分离株DNT粗提原液及10和100倍稀释物分别注射于4~5、15~20和40~45 d 3组不同日龄的SPF鼠,进行毒力检测。结果分离到一株支气管败血波氏杆菌,其DNT毒力较强,0.2 mL粗提原液可造成4~5 d乳鼠皮下组织发生严重急性出血性炎症而死亡;15~20 d乳鼠皮下出血,毛囊损伤,表皮层细胞结构模糊、紊乱,耐过乳鼠损伤处皮肤毛发生长受损;40~45 d小鼠脱毛等轻微症状。本试验分离菌株皮肤毒性较强,DNT对不同日龄鼠的皮肤致病力有差异,可引起表皮层、真皮层和皮下组织同时损伤。

Isolation and identification of Bordetella bronchiseptica from swine and skin virulence test of DNT crude extraction to mice

Abstract:
To prevent Bordetella caused disease better, a sample of swine nose swab with obvious respiratory disease from certain pig farm in Sichuan was authenticated by cultivation, biochemical test, PCR and homology analysis.To investigate the virulence of dermonecrotic toxin (DNT), DNT crude extraction obtained by the classical method including 10 and 100 times dilutions of DNT crude extraction, were injected into the 4-5、15-20 and 40-45 d SPF mice, respectively.One B.bronchiseptica strain was separated successfully and its DNT virulence was high.0.2 mL original crude extraction caused acute haemorrhagic inflammation and death to 4-5 d suckling mice;And the 15-20 d suckling mice got injured in hair follicle, and hemorrhage in subcutaneous tissue, the layer of cell structure was in disorder and the hair growth of injured skin was influenced while 40-45 d mice only displayed unhairing symptom.In conclusion, this strain has high virulence, and its DNT can cause varying degrees of injury to the mice at different days old, and lead to injuries in Epithelium, corium layer and subcutaneous tissue.The study provided some reference for the prevention of B.bronchiseptica.
