为探究侧脑室注射Nesfatin-1对雌性大鼠促性腺激素水平及其mRNA表达的影响,对初情期前雌性大鼠的侧脑室注射Nesfatin-1,采用实时荧光定量PCR法检测促黄体素(Luteinizing hormone,LH)和促卵泡素(Follicle-stimulating hormone,FSH) mRNA在垂体中的变化,采用酶联免疫分析法检测血清LH和FSH的浓度.结果表明:在初情期前的雌性大鼠中,Nesfatin-1可促进初情期启动、增加卵巢重量、血清LH水平、垂体LH mRNA和FSH mRNA的表达增加(P< 0.05);但Nesfatin-1对FSH水平的影响不突出,注射后15 min时增加(P< 0.05),60 min 不产生影响(P> 0.05).侧脑室注射Nesfatin-1后15和60 min均能提高血清LH 水平,15 min时提高更明显(P< 0.05),但对FSH水平的影响不尽相同.在成年的雌性大鼠中,Nesfatin-1对LH和FSH的分泌不产生影响(P> 0.05).Nesfatin-1可通过下丘脑诱导和增强初情期过渡阶段雌性大鼠LH和FSH 的释放及LH、FSH mRNA的表达. |

Effect of nesfatin-1 on the level of LH, FSH and the genes expression in female rats |

To study the effect of nesfatin-1 on gonadotropin secretion and their mRNA expression by intracerebroventricular injection at the central level in rats.Female rats at pre-puberty were treated with nesfatin-1 by intracerebroventricular injection.The luteinizing hormone (LH) and follicle-stimulating hormone (FSH) mRNA expression in pituitary were determined with real-time quantitative PCR.The concentration of serum LH and FSH were determined with enzyme-linked immunosorbent (ELISA).The results showed that,nesfatin-1 could significantly advance vaginal opening,enhance ovarian weight,serum LH level and mRNA expression of LH,FSH in pituitary during the pre-puberty (P< 0.05);but FSH level was significantly increased at 15 minutes after intracerebroventricular injection (P< 0.05) but not at 60 minutes after intracerebroventricular injection (P> 0.05).Serum LH level was increased at 15 and 60 minutes after intracerebroventricular injection of nesfatin-1,where LH level was increased more at 15 min compared to 60 min (P< 0.05).Nesfatin-1 could not affect the secretion of gonadotropin in adult rats (P> 0.05).Nesfatin-1 could induce and enhance the secretion and mRNA expression of FSH and LH through the hypothalamus during the pubertal transition. |
