制动应激对妊娠早期孕鼠小肠黏膜组织结构的影响

摘要:
通过观察应激状态下孕早期母鼠摄食量、体重以及小肠组织学结构的变化,探究应激对孕鼠消化能力的影响,以期揭示应激对妊娠的影响机制。以妊娠早期孕鼠为研究对象,束缚制动方法建立制动应激模型,每天1次,每次4 h,于妊娠3、5和7 d取材,记录食物摄取量和体重变化,应用PAS染色和PCNA免疫组织化学染色,观察应激对孕鼠妊娠早期十二指肠、空肠、回肠杯状细胞及上皮细胞增殖的影响。结果发现制动应激组孕鼠食物摄取量和体重均出现显著下降,在 E3d、E5d和E7d食物摄取量分别下降了15%、11%和19%,体重分别下降了71%、90%和105%;应激组小鼠十二指肠、空肠、回肠肠绒毛高度、杯状细胞数量和肠腺上皮细胞中的PCNA阳性细胞率均出现显著降低。制动应激显著抑制孕鼠的肠黏膜组织生长发育,可能由此降低孕鼠的消化能力,从而不利于妊娠发生。

Effects of restraint stress on the histological structure of small intestinal mucosa in early pregnant mice

Abstract:
The research focuses on assessing the impact of restraint stress on the digestive capacity of early pregnant mice and revealing the effect mechanism of stress on pregnancy by observing the changes of the food intake,body weight and small intestinal mucosa histological structure in the pregnant mice with restraint stress.The early pregnant mice were treated by restraint stress continuously for 4 hours per day till being killed.The mice were sacrificed to isolate the small intestine at pregnancy of 3,5 and 7 days (named as E3d,E5d and E7d).The changes of food intake and body weight were recorded,and PAS staining and immune histochemical staining of PCNA were used to observe the effect of stress on the goblet cells and epithelial cell proliferation in duodenum,jejunum,ileum of early pregnant mice.The food intake and body weight in stress mice dropped significantly.The food intake had fallen by 15%,11% and 19% at E3d,E5d and E7d respectively,and the weight had fallen by 71%,90% and 105% respectively.The restraint stress could reduce the intestinal goblet cells numbers,villous height,and the rate of PCNA positive cells expression in duodenum,jejunum and ileum of mice.Stress significantly inhibits the growth of the intestinal mucosa and may thus reduce the digestion ability of the pregnancy mice,which is harmful to pregnancy.
