果胶酶和纤维素酶参与管花肉苁蓉侵入寄主根的研究

摘要:
采用3, 5-二硝基水杨酸(3, 5-dinitrosalicylic acid, DNS)比色法和超微细胞化学定位法, 研究了果胶酶和纤维素酶在管花肉苁蓉芽管、吸器和侵染结构界面的分泌和分布及对寄主根细胞的作用。结果表明, 离体的芽管、吸器均分泌果胶酶和纤维素酶, 其含量随着芽管和吸器生长呈增加趋势。在吸器和寄主根的侵染结构界面, 侵入细胞也分泌果胶酶和纤维素酶, 并在吸器和寄主根相邻的胞间层、寄主细胞壁周边部位分布, 导致寄主细胞间连接被解除和细胞的降解。结论:管花肉苁蓉吸器侵入寄主根的过程中有果胶酶和纤维素酶的参与。

Pectinase and cellulase produced by the parasite plant Cistanche tubulosa are involved in parasitising its host root

Abstract:
The secretion and distribution as well as effects on host cells of pectinase and cellulase were studied in germ tubes, haustorium and the host-parasite interface of Cistanche tubulosa by 3, 5-dinitrosalicylic acid (DNS) method and ultracytochemical localization.It was shown that pectinase and cellulase were secreted from in vitro germ tube and haustorium.The activities of pectinase and cellulase were increased with growth of germ tube and haustorium.In the host-parasite interface, pectinase and cellulase were secreted from invasive cell of haustorium, then distributed to intercellular layer of haustorium-host root interface and host cells.The functions of pectinase and cellulase were to damage host cells connection and structures for the purpose of haustorium invasion.This is the first report that pectinase and cellulase are involve in invasion of C.tubulosa haustorium to the root of Tamarix chinesis.
