耕层柽柳根生长分布和管花肉苁蓉接种的WINRHIZO扫描观察

摘要:
为了研究耕层土壤中柽柳根和管花肉苁蓉的相互作用,明确管花肉苁蓉的接种时间、生长和药用成分动态,采用WINRHIZO根系扫描系统,定期检测柽柳根系生长和肉苁蓉接种;采用HPLC和分光光度法测定松果菊苷、毛蕊花糖苷和可溶性糖含量。结果表明:柽柳茎段在扦插后15 d生根,根的生长遵循"S"曲线规律;首次观察到管花肉苁蓉与柽柳根建立稳定寄生关系的时间为扦插后60 d,管花肉苁蓉的接种率动态变化遵循"S"曲线规律;管花肉苁蓉单株干重在接种后90 d内增长缓慢,90 d后快速增长,至150 d单株干重达16 g;生长150 d的管花肉苁蓉含松果菊苷9.6%,毛蕊花糖苷1.7%,苯乙醇苷总量11.3%,可溶性糖0.19%。WINRHIZO扫描分析为确定耕层土壤中寄生植物和寄主的接种动态及影响因素研究提供了新方法。

Monitor of tamarisk root growth and Cistanche tubulosa infection in soil arable layer by WINRHIZO

Abstract:
In order to study the interaction of tamarisk root and Cistanche tubulosa in the soil arable layer,we used WINRHIZO scanner system to determine the tamarisk root growth and detect the time when the C.tubulosa would infect the host root;we measured the echinacoside,verbascoside and soluble sugar of the C.tubulosa by HPLC and spectrophotometry.The result showed:the tamarisk stem cuttings began to develop the root 15 days after cuttage ,and the growth curve followed "S" curve;the C.tubulosa began to infect with tamarisk on day 60 after cuttage for the first time.Similarly,the infection rate of the C.tubulosa followed "S" curve;initially,the growth of C.tubulosa plant grew slower during the 90 days of infection (indicated by lighter dry weight),then the C.tubulosa began to grow faster after the day 90,and the dry weight of the single plant could reach to as high as 16 gram,and the amount of the echinacoside,verbascoside and soluble sugar were 9.6%,1.7% and 11.3% separately.Our study indicated that the WINRHIZO scanner system could be be used as a new approach to detect the parasite plant and the host plant in the soil arable layer and provide more detailed information on the infection dynamics and the effective factors during the complicated infection processes.
