獐茅液泡膜Na+/H+逆向转运蛋白基因AlNHX转化水稻的耐盐性研究

摘要:
为提高转基因植物的耐盐性,本研究利用农杆菌介导的方法将獐茅液泡膜Na+/H+逆向转运蛋白基因AlNHX组成型表达在日本晴粳稻中。PCR鉴定结果表明,AlNHX基因已经整合到日本晴粳稻中,并在转录水平上表达。转AlNHX基因水稻的耐盐性明显提高。用NaCl经5 d培养处理后,转基因水稻可以在含有0~200 mmol/L NaCl的MS培养基中生长,而野生型水稻无法在含盐量超过150 mmol/L NaCl的MS培养基中生长。

Effects of transformating a vacuolar Na+/H+ antiporter gene from Aeluropus littoralis on salt-tolerance in rice

Abstract:
Establishment of high efficiency Agrobacterium-mediated transgenic techniques has greatly accelerated the widespread application of transgene in rice.It was very important to improve the salt-tolerance.In this study,the ORF of vacuolar Na+/H+ antiporter gene was transferred into nipponbare via Agrobacterium-madiated method.The transformed plants were verified by PCR,over expression of AlNHX in rice conferred high salt tolerance to the transgenic plants.The transgenic plants could survive in the medium containing up to 200 mmol/L NaCl for 15 days,while the wild-type plants failed to grow higher than 150 mmol/L NaCl.
