‘峰后’葡萄果实发育与赤霉素(GA3)处理以及VvSEP3基因表达的关系

摘要:
为了探明‘峰后’葡萄赤霉素(GA3)处理后促使果粒变长的原因和VvSEP3基因在果实早期发育中的作用,在开花前利用0.1 mg/g GA3处理花序并通过原位杂交分析了VvSEP3基因的表达。结果显示:GA3处理提高了峰后葡萄的坐果,增加了果形指数,从开花后14 d开始到开花后56 d,GA3处理后的果实纵径始终大于同时期的对照果实,解剖学结构研究表明,中果皮的细胞膨大从开花后7 d开始明显大于同时期发育的对照果粒。VvSEP3基因在开花期的柱头、花柱、横膈膜以及胚珠的珠心组织中有表达,开花后主要在子房壁中表达,当幼胚发育时在维管束组织和胚乳组织中有所表达。本研究显示GA3处理后明显增加峰后葡萄的果形指数,同时促进了果肉细胞的纵向生长从而使果粒变长;VvSEP3基因在子房壁细胞分裂与膨大过程中可能发挥重要作用,同时也可能参与了种子的形成和发育过程。

Berry development in relation to Gibberellin(GA3) treatment andVvSEP3 expression of ‘Fenghou’ grapevine

Abstract:
This paper is concerned with The effects of gibberellins (GA3) on berry growth and the possible role of VvSEP3 gene in early berry development of ‘Fenghou’ grapevine (Vitis vinifera L.).The inflorescences before flowering were treated with 0.1mg/L GA3.The VvSEP3 gene expression pattern during early berry development was studied by in situ hybridization.Our results showed that GA3 treatment increased the fruit setting rate and the fruit form index.The longitudinal diameter of GA3 treatment berry was larger than the control from 14 days after full bloom (DAFB) to 56 DAFB.Anatomical structure of the berry showed that the cell longitudinal diameter of the mesocarp was significantly larger than control berry from 7 DAFB,indicating that berry elongation due to GA3 treatment might be the stimulation of cell vertical growth.The VvSEP3 gene was expressed in the stigma,style,diaphragm and the nucellar tissue of ovules during flowering,in the ovary wall after flowering,and in vascular tissues and endosperm tissues during young embryo development.These results suggested that the VvSEP3 gene might play a role in the ovary wall cell division and enlargement process,and also involved in the process of seed formation and development.
