棉花盐胁迫应答基因GhCPK5的克隆及序列分析

摘要:
本研究克隆了1个陆地棉新的钙依赖蛋白激酶基因GhCPK5(Genbank登录号为:HM002634)。GhCPK5基因组DNA序列全长3 031 bp,包含一个1 707 bp的开放阅读框,具有7个外显子和6个内含子。GhCPK5可能定位于细胞核内,有568个氨基酸残基,分子量为63 ku。氨基酸序列分析表明,陆地棉GhCPK5与杨树PtCPK5同源性达到89%,同样包含完整的激酶结构域和4个EF-hand结构域。RT-PCR分析表明,该基因在根、茎、叶等组织中表达;在盐胁迫处理下,GhCPK5在各组织的表达量明显上升。

Isolation and sequence analysis of salt responsive geneGhCPK5 from Gossypium hirsutum L.

Abstract:
In this study,a salt responsive gene GhCPK5 was firstly obtained from upland cotton using RT-PCR and RACE.GhCPK5 located probably in cell nucleus and the genomic sequence indicated that GhCPK5 had 7 exons and 6 introns.GhCPK5 contained an open reading frame (ORF) of 1 707 bp,which encoded a protein with 568 amino acids,with a molecular weight of 63 ku.GhCPK5 contained one protein kinase domain and four conserved EF-hand domains.The amino acid sequence of GhCPK5 shared 89% identify with Populus trichocarpa homolog PtCPK5.RT-PCR analysis revealed that GhCPK5 expressed in root, stem and leaf, and its expression level was significantly up-regulated by salt stress.
