无刺桅杆槐和无刺槐再生体系的研究

摘要:
以无刺桅杆槐和无刺槐的带腋芽茎段为材料获取无菌苗,再以无菌苗叶片为外植体进行植株再生研究。结果表明:2种无刺槐茎段的最佳取材时间均为5月上旬;无刺桅杆槐适宜的叶片愈伤组织诱导培养基为MS+6-BA 0.25 mg/L+2,4-D 6.00 mg/L,无刺槐为MS +6-BA 0.50 mg/L+2,4-D 2.00 mg/L,愈伤诱导率分别为60%、80%;不定芽诱导培养基,无刺桅杆槐和无刺槐均为MS+6-BA 5.00 mg/L+IBA 0.10 mg/L+GA3 0.10 mg/L,再生率分别为60%、50%。2种无刺槐的生根培养基均为1/2MS+IBA 0.05 mg/L,生根率分别为55.6%、72.2%,组培苗移栽成活率达85%以上。

Regeneration for Robinia pseudoacacia var.Inermis andRobinia pseudoacacia var.rectissima 'inermis’

Abstract:
Aseptic seeding were cultured using stems with bud as materials which coming from two varieties of no-barbed Robinia.Take leaves of aseptic seeding as explants for researching regeneration.Results showed that the best time of Robinia pseudoacacia var.Inermis to take stems was in early May as same as Robinia pseudoacacia var.rectissima 'inermis’.The suitable callus medium was different for the two varieties,Robinia pseudoacacia var.rectissima 'inermis’ was MS+6-BA 0.25 mg/L+2,4-D 6.00 mg/L,Robinia pseudoacacia var.Inermis was MS+6-BA 0.50 mg/L+2,4-D 2.00 mg/L,the callus initiation rate were 60% and 80%;Proper culture medium for Robinia pseudoacacia var.rectissima 'inermis’ and Robinia pseudoacacia var.Inermis was MS+6-BA 5.00 mg/L+IBA 0.10 mg/L+GA3 0.10 mg/L,regeneration rate were 60% and 50%;Two kinds of varieties in this paper had same medium for rootage culture which was 1/2MS+IBA 0.05 mg/L,but the rootage rate was different,Robinia pseudoacacia var.rectissima 'inermis’ was 55.6%,another is 72.2%,the survival rate of transplant for tissue culture seedings was more than 85%.
