奶牛初乳与常乳乳蛋白变化的比较蛋白质组学研究

摘要:
为分析初乳与常乳乳蛋白的表达变化,揭示乳蛋白分泌及乳腺易感疾病的机理,采用二维凝胶电泳结合液相色谱串联质谱的方法分析了产后第1天、第7天和第21天牛乳中蛋白表达变化。结果发现,相对于第7天和第21天牛乳蛋白的表达丰度,第1天牛乳蛋白中有5个蛋白表达量增加;而第7天与第21天牛乳蛋白表达无变化。且第1天牛乳蛋白表达量增加的是具有免疫活性的免疫球蛋白M和G以及运输功能的白蛋白和转铁蛋白等。结果表明由初乳向常乳转换过程中,这些乳蛋白表达量的降低可能与乳腺易感乳房炎等疾病相关。

Developmental changes of the milk protein from colostrumsto milk in the periparturient dairy cattle

Abstract:
The purpose of this study was to analyze the changes of milk protein from colostrum to milk, and further insight into the mechanisms of milk protein secretion and defense of the mammary gland. Milk proteins from the dairy cows at 1, 7 and 21 days after calving were separated and identified by two-dimensional electrophoresis in combination with HPLC tandem ion trap spectrometry. The results showed that five proteins in the first day were up-regulated compared with the 7 and 21 d after calving, but there was no different between 7 and 21 d. In addition, highest proteins expression in colostrum, including the immune function proteins IgM, IgG, transport proteins transferrin and albumin, were identified. These results indicated decreased proteins expression in colostrum might be related to mastitis in the periparturient dairy cattle.
