红豆草体细胞胚胎发生系统的初步建立

摘要:
采用红豆草(Onobrychis viciaefolia Scop)6个栽培品种无菌苗下胚轴切段,在6-BA和 IAA 不同浓度配比的 MS 培养基上诱导愈伤组织。将其在 MS 附加一定浓度2,4-D 的培养基上进行多次继代培养,通过筛选、克隆,从6个品种的愈伤组织中获得了大量的胚状体,诱导率最高达100%。定期将上述愈伤组织进行更新及继代培养达1年,仍能保持较高程度的体细胞胚胎发生能力。将含有胚状体的愈伤组织转入无激素的 MS 培养基上后,在3个品种中,均不同程度地分化出带根的完整健康小植株及一些畸形株。

Abstract:
Calli were formed from hypocotyl explants of six cultivars of sainfoin(Onobrychis viciaefolia Scop.)cultured on MS medium containing different concentrations of 6-BA and IAA.The calli were subcultured on MS medium supplemented with 2,4-D.Through screening and cloning,a num- ber of somatic embryos were obtained from each of the six cultivars and the induction rate was as high as 100%.After one year of subcultures,the calli had still been keeping a high devoloping ability of somatic embryogenesis.When they we...
