芸苔属两个种原生质体的分离和培养

摘要:
由甘蓝(Brassica oleracea)和芜菁(Brassica rapa)黄化小苗子叶和下胚轴分离出大量有活力的原生质体,并成功地进行了培养。30小时后发生第一次分裂。下胚轴原生质体分裂频率明显高于子叶原生质体。除芜菁子叶原生质体外,其余材料的原生质体均能持续分裂,20天后形成肉眼可见的愈伤组织。在 MS 培养基上,来源于甘蓝子叶原生质体的愈伤组织分化出根,其余的未见分化现象。

Abstract:
A Iarge number of viable protoplasts were isolated by enzyme treat- ment from cotyledons and hypocotyls of etiolated seedlings of Brassica oleracea and Brassica rapa,and were cultured successfully.The first division was observed after 30 hours.The division frequency of hypoco- tyl protoplasts was obviously higher than that of cotyledon protoplasts.
